Effect of Different Permeability Enhancers and Effect of PH in Atropine Sulphate Sublingual Tablets

write a little literature review about the Atropine Sulphate and its use in Organophosphorous poisoning which you can found in the resource and you can use an extra if you need, and about the enhancers that I am going to use which are: Sodium Dodecyl Sulphate, Chitosan, and Palmitoyl-L-carnitine chloride. There is an article for all of them in the references.
So, I am making the same formula my senior used but I add those enhncers to them one by one and study the effect of them. I do a quality control studies which include: Content uniformity test, Dissolution test, Disintegration test, Friability test, Force Test, Diffusion test. The other step is studying the effect of the PH to the AS without the enhancers. I want a need statement (why this is important ) and an objective and goal which is divided in two part studying the permeability enhancers and their effect on AS . and studying the effect of PH in AS to see which PH is the best for better permeability. And why is that important which is Sublingual are easily taken than AS injector in the market.

In the method section I want first the Material section which include:
Atropine Sulphae (Sigma, St. Louis, MO)
Chitosan (Sigma, St. Louis, MO)
Palmitoyl-L-carnitine chloride (Sigma, St. Louis, MO)
Sodium Dodecyl Sulohate (Sigma, St. Louis, MO)

Then method which includes all the tests I listed previously:
Friability Test:
Take 120 tablets, weigh them, put them in the friability tester for 100 round with speed 25 rpm. After finishing weigh them again and calculate the % loss
Content Uniformity Test:
Take 10 tablets, weigh them individually, put every tablet in a tube and dilute it with 10 ml water, take a small amount of the solution after filter it and analyze it by HPLC
Dissolution Test:
Take 12 tablets put every tablet in a filter flask with 2ml water and 6 tablets turn the vaccum on after 60 sec and 6 tablets after 120 sec. After that take the 2 ml and analyze it by HPLC
Force Test:
Take 10 tablets measure their weight, diameter, thickness, and hardness by hardness tester.
Disintegration test:
We do disintegration in two temp medium: 25oC water, and 37oC water. In 3 speed 40 rpm, 60rpm, and 80rpm. Using two volume 2 ml and 20 ml
Diffusion permeability test:
It is the most important step we use a porcine sublingual membrane cut surgically. We use the franz cells in order to measure the diffusion the receptor chamber is filled with Phosphate Buffer and the membrane placed in place . fill the donor chamber with the buffer and place your tablet then after specific time withdraw from the receptor chamber 1 ml and place it in HPLC vial to analyze it, at the same time place 1 ml of the buffer in the receptor chamber. Repeat the steps till the end of time sheet.

However in the PH study we are just going to do the diffusion test as I describe using Mcilvaine’s Buffer (A citrate/phosphate buffer) instead of Phospahe buffer and using AS sublingual tablet without any enhancer.
